Correlation A comparison of this method using an automated analyzer was carried out against a reference method based on the ferene reaction. The performance of Catachem’s UIBC method was found essentially equivalent to the reference method used. Linear regression analysis produced the following statistics: |

Reagent Storage and Stability Store the Catachem UIBC Color Reagent at 2-8°C. Store the Catachem UIBC Buffer and Iron Standard at 18-26°C. When stored as directed, the reagents are stable until the expiration date stated on the label
Reagent Preparation The Catachem UIBC method requires two Working Reagents, R1 and R2. R2 is packaged ready for use. No preparation is required. The Working UIBC Buffer (R1) is prepared by adding the contents of the iron standard to the UIBC buffer.
Method Performance Characteristics Sensitivity: The sensitivity of this method is 0.00025-0.00030 absorbance units per ug/DL. Linear Range: In this method there is no significant nonlinearity over the range of 0 - 500 ug/dL. Precision: Within-run and day-to-day precision is summarized below. |

Intended Use For IN VITRO diagnostic use in the quantitative determination of Unsaturated Total Iron Binding Capacity (UIBC) in serum using manual or automated applications.
Method Principle A buffer system at alkaline pH is used to measure UIBC. Excess ferrous ions bind specifically to the serum sample transferrin protein unfilled iron-binding sites the remaining unbound ferrous ions are measured using the total iron ferene reaction. The difference between the amount of unbound iron and the total amount of iron added to the serum sample is equivalent to the amount bound to the transferrin protein molecule. This amount of bound iron is the UIBC. The serum TIBC is equal to the total iron and the UIBC. The reaction scheme illustrates the reaction that occurs in the method. |
